A professional dancer who trained with the American Ballet Theatre and other prestigious companies, Thompson became a familiar face through Burger King ads of the 1980s. She landed her first film role in All the Right Moves (1983), as Tom Cruise's girlfriend. Thompson was memorable as Michael J. Fox's mom (made up to look older) in Back to the Future (1985) and its sequels, and has also appeared in Some Kind of Wonderful (1987) and Dennis the Menace (1993). She starred in her own sitcom, Caroline in the City (1995–1999) and in the Lifetime network drama For the People (2002–2003). She plays a secret agent/housewife in the Hallmark Channel's Jane Doe mystery movies series (2005–).
